Drug intelligence / Profile preview
[68Ga]Ga-DOTA-RET-L7 is a preclinical, research-stage radiopharmaceutical developed as a positron emission tomography (PET) imaging agent. It consists of the RET-L7 peptide, which was discovered via phage display and selected for its high-affinity binding to the receptor tyrosine kinase RET, conjugated to the chelator DOTA and radiolabeled with gallium-68 (68Ga). RET is highly overexpressed in neuroendocrine prostate cancer (NEPC), an aggressive, treatment-resistant subtype that is frequently prostate-specific membrane antigen (PSMA)-low or PSMA-negative. In preclinical mouse xenograft models, [68Ga]Ga-DOTA-RET-L7 demonstrated high, specific tumor uptake in RET-positive NEPC tumors with rapid blood clearance and strong self-blockade. It forms a theranostic pair with its therapeutic partner, [177Lu]Lu-DOTA-RET-L7, offering a promising diagnostic and therapeutic approach for PSMA-negative prostate cancer.
